摘要: 溶酶体功能障碍是AD病理机制的重要一环,其功能修复为疾病干预提供新契机。本文系统综述了溶酶体功能恢复的六大策略:转录因子EB的激活与调控、自噬-溶酶体途径的调节、中医药干预、运动疗法、纳米材料的应用及新型溶酶体增强剂的开发,重点探讨其分子机制、模型验证及临床转化潜力。通过整合基础研究与转化医学进展,本文旨在为AD治疗提供理论依据与创新方向。

Abstract: Lysosomal dysfunction is an important part of the pathological mechanism of Alzheimer’s disease, and its functional repair provides a new opportunity for disease intervention. This paper systematically reviews six major strategies for restoring lysosomal function: activation and regulation of transcription factor EB, modulation of the autophagy-lysosome pathway, traditional Chinese medicine interventions, exercise therapy, application of nanomaterials, and the development of novel lysosomal enhancers. It also provides an in-depth discussion of the molecular mechanisms, model validation, and clinical translation potential. By integrating basic research and advances in translational medicine, this review aims to provide a theoretical basis and innovative direction for Alzheimer’s disease therapy.
