Abstract: Hearing impairment, one of the most prevalent age-related syndromes, ranks as the third leading health condition affecting quality of life in the eldery. Its complex pathogenesis results from the combined effects of genetic, environmental, and metabolic factors. While single-omics approaches are limitated in comprehensively analyzing this intricate mechanism, the recent emergence of multi-omics technologies has opened new avenues for revealing the pathogenic mechanisms of hearing disorders. This review synthesizes recent advancements in genomics, epigenomics, transcriptomics, proteomics, metabolomics, and multi-omics integration within the field of hearing impairment research. Although multi-omics studies have deepened our understanding of hearing disorder mechanisms, future research must focus on mechanistic exploration and clinical translation to advance precision prevention and treatment strategies.
