黄芪多糖对老年晚期胃癌病人外周血中调节性T细胞的影响

Effects of astragalus polysaccharide on expression and function of regulatory T cells in elderly patients with advanced gastric cancer

摘要: 目的 探讨黄芪多糖对老年晚期胃癌病人外周血中调节性T细胞(regulatory T cells, Tregs)表达及功能的影响。方法 收集60例老年晚期胃癌病人,随机平均分为对照组(5%葡萄糖注射液)、低剂量组(黄芪多糖125 mg)及高剂量组(黄芪多糖250 mg),各组均于用药前后,应用流式细胞仪检测病人外周血中CD4+CD25+Tregs及CD4+CD25+FoxP3+Tregs的表达,应用ELISA法检测病人外周血中Treg相关抗炎细胞因子IL-10和转化生长因子-β1(TGF-β1)的含量,比较2组用药前后的差异。结果 高剂量组外周血CD4+CD25+Tregs及CD4+CD25+FoxP3+Tregs的表达较用药前均明显降低(P<0.05),而低剂量组及对照组未见明显减少。同时高剂量组的IL-10及TGF-β1水平较用药前明显减少(P<0.05);而对照组的IL-10及TGF-β1水平较用药前未见明显减少,低剂量组TGF-β1水平较用药前降低(P<0.05)。结论 黄芪多糖可能通过降低Tregs的表达及功能减轻老年晚期胃癌病人的免疫抑制状态。

Abstract: Objective To investigate the effect of astragalus polysaccharide on regulatory T cells (Tregs) in the peripheral blood of the elderly patients with advanced gastric cancer. Methods A total of 60 elderly patients with advanced gastric cancer were randomly divided into control group, low-dose astragalus polysaccharides group and high-dose astragalus polysaccharides group. Flow cytometry was used to detect the expression of CD4+CD25+Tregs and CD4+CD25+FoxP3+Tregs in the peripheral blood before or after the treatment. The levels of interleukin-10 (IL-10) and transforming growth factor-β1(TGF-β1) were detected by enzyme linked immunosorbent assay method. Results The expressions of CD4+CD25+Tregs and CD4+CD25+FoxP3+Tregs in the peripheral blood of the high-dose group were significantly lower than those before the administration (P<0.05).However, the expressions of CD4+CD25+Tregs and CD4+CD25+FoxP3+Tregs were not significantly decreased in the low-dose group and the control group. At the same time, the levels of IL-10 and TGF-β1 in the high-dose group were significantly lower than those before the administration (P<0.05), while showing no significant changes in the control group. The level of TGF-β1 in the low-dose group was lower than that before administration(P<0.05). Conclusions Astragalus polysaccharide may reduce the immunosuppressive status of the elderly patients with advanced gastric cancer by reducing the expression and function of Tregs.
