针刺干预对绝经后骨质疏松症模型大鼠肠道菌群影响的研究

Effect of acupuncture intervention on intestinal microflora of postmenopausal osteoporosis model rats

摘要: 目的 观察绝经后骨质疏松症模型大鼠经电针干预后与骨代谢密切相关的肠道菌群丰度的变化。方法 选取32只3月龄雌性SD清洁级大鼠,其中8只为空白组,8只为假手术组,16只行双侧卵巢摘除术后随机平均分为2组,分别为模型组和电针组。电针组取穴:(1)关元、三阴交(双);(2)肾俞、后三里(双)。两组穴位交替进行,每次电针刺激20 min,每日1次,每周连续干预5 d,共12周。其余3组仅常规饲养,不作特殊处理。12周后处死大鼠,应用骨密度仪统一测定右侧胫骨、股骨的骨密度,采用16S rRNA基因测序法测量大鼠粪便中肠道菌群结构变化。 结果 各组大鼠骨密度的变化:与空白组、假手术组比较,模型组大鼠骨密度明显降低(P<0.05);与模型组比较,电针组骨密度明显提高(P<0.05)。各组大鼠肠道菌群的变化:在门水平,与空白组、假手术组相比,模型组的软壁菌门、放线菌门、脱铁杆菌门丰度增加(P<0.05),厚壁菌门、拟杆菌门、变形菌门、 黑水仙菌门丰度降低(P<0.05);与模型组相比,电针组的厚壁菌门、拟杆菌门、变形菌门、黑水仙菌门丰度增加(P<0.05),软壁菌门、放线菌门、脱铁杆菌门丰度降低(P<0.05)。在属水平,与空白组、假手术组相比,模型组拟杆菌属、乳杆菌属、肠球菌属丰度降低(P<0.05);与模型组相比,电针组拟杆菌属、乳杆菌属、肠球菌属丰度增加。结论 电针干预可以提高去卵巢模型大鼠的骨密度,调节去卵巢模型大鼠的肠道菌群结构。

Abstract: Objective To observe the changes of intestinal flora related to bone metabolism in the postmenopausal osteoporosis model rats receiving the intervention of electroacupuncture. Methods A total of 32 3-month-old female SD rats were randomly divided into blank group, sham operation group, model group and electroacupuncture group, with 8 rats in each group. The rats in the model group and the electroacupuncture group underwent bilateral ovariectomy. The electroacupuncture group was alternately given electroacupuncture intervention at "Guanyuan(CV4) + Sanyinjiao(SP6)"(double) and "Shenshu(BL23) + Housanli(ST36)"(double) for 20 min each time, once a day, for 5 consecutive days every week for 12 weeks. After 12 weeks, the bone mineral density(BMD) of the right tibia and femur was measured by bone densitometer. The feces were taken for 16S rDNA flora detection. Results Compared with blank group and sham operation group, the level of BMD in model group was significantly decreased(P<0.05). Compared with model group, the level of BMD in electroacupuncture group was significantly increased(P<0.05). At the phylum level, the abundance of Tenericutes, Actinobacteria and Deferribacteria in model group increased significantly compared with blank group and sham operation group(P<0.05), while the abundance of Firmicutes, Bacteroidetes, Proteobacteria and Melainabacteria decreased(P<0.05). Compared with the model group, the abundance of Firmicutes, Bacteroidetes, Proteobacteria and Melainabacteria in electroacupuncture group increased(P<0.05), and the abundance of Tenericutes, Actinobacteria and Deferribacteria in electroacupuncture group decreased(P<0.05). At the genus level, the abundance of Bacteroides, Lactobacillus and Enterococcus in model group decreased(P<0.05) compared with blank group and sham operation group. Compared with model group, the abundance of Bacteroides, Lactobacillus and Enterococcus in electroacupuncture group increased. Conclusions Electroacupuncture can improve the level of BMD of ovariectomized rats and regulate their intestinal microflora structure.
