人乳头瘤病毒致宫颈癌细胞免疫逃逸机制探讨

摘要:

目的探讨人乳头瘤病毒(HPV)感染与宫颈癌细胞免疫逃逸之间的关系,揭示HPV在宫颈癌发生、发展中的作用。方法以宫颈癌细胞系,宫颈癌组织及其相应的正常宫颈组织为材料,采用实时聚合酶链反应(PCR)检测HPV mRNA表达水平,采用流式细胞仪进行宫颈癌细胞表面C3b沉积检测。结果在30对配对组织中有26对肿瘤组织HPV表达高于配对的癌旁正常组织,C3b的沉积在HPV感染宫颈癌细胞组明显低于无感染组,尤以老年组变化更为明显,同时宫颈癌细胞的迁徙和侵袭能力在HPV感染组明显升高。结论HPV感染的宫颈癌组织通过抑制C3b的沉积而引发免疫逃逸,在宫颈癌的发生、发展中发挥重要作用。

Abstract:

ObjectiveTo investigate the effect of HPV infection on immune escape of cervical cancer cells.MethodsThe level of HPV mRNA was explored by realtime PCR, meanwhile, the deposition of C3b was detected by flow cytometry.ResultsThe levels of HPV mRNA and C3b deposition were significantly increased in human cervical cancer tissue, especially in elderly group, which contributed to the migration and intrusion of cervical cancer cells.ConclusionsThe results indicate that the infection of HPV might involve in migration and intrusion of cervical cancer cells through inhibiting the deposition of C3b.
